魚類急毒性替代測試方法與整合型測試評估方法建立(1/2)
中文摘要 | 隨著人口增長,人類的日常活動釋放的污染物對水體環境造成潛在危害,影響生態系統和人類健康。目前環境風險危害評估主要依賴於大量動物試驗的生態毒理學數據,對動物福祉造成負面影響。儘管替代測試方法的使用是一項全球趨勢,但單一替代測試往往無法完全取代動物試驗數據。因此,國際上正在推動整合型測試評估方法(Integrated approaches to testing and assessment, IATA),以整合多種數據協助制訂政策決策,從而保護環境安全。本計畫旨在建立魚鰓細胞株急毒性檢測方法,發展魚類急毒性IATA,並收集新興替代測試方法資訊。本計畫之5次參考毒物試驗顯示的劑量反應符合標準,並且以10項與放流水相關之物質評估魚鰓細胞株試驗之適用性,結果顯示細胞株試驗與魚類試驗兩者間存在顯著高度相關,r值為0.7732(p < 0.05)。雖然大多數物質在細胞株試驗中靈敏度低,但66.7%的物質在細胞株試驗的EC50與魚類急毒性試驗的LC50差異小於10倍,於本年度將研究結果與國際規範彙整後撰寫出魚鰓細胞株試驗標準方法草案一式。另外本計畫以10項物質評估各替代試驗及IATA的相關性與靈敏性。結果證實不論是替代測試或是IATA,都跟魚類急毒性試驗之間具有高度相關,受到閾值濃度影響,IATA預測魚類急毒性結論大多高估物質毒性,能在盡可能減少動物實驗的前提下,幫助篩選可能有毒之物質,與達到保護環境之效能。綜上所述,虹鱒魚鰓細胞株試驗作為替代測試方法,展現出良好的預測潛力。IATA預測結果有助於減少對物質毒性的低估,面對其發展挑戰,建議積極採用新興替代測試方法,以促進環境及人類健康風險的評估與管理。 | ||

Alternative Testing Methods for Fish Acute Toxicity and the Development of Integrated Approaches to Testing and Assessment (1/2)
英文摘要 | With population growth, pollutants released from human daily activities pose potential hazards to the aquatic environment, impacting ecosystems and human health. Currently, environmental risk assessments mainly rely on ecotoxicological data derived from extensive animal testing, which negatively affects animal welfare. Although using alternative testing methods has become a global trend, individual alternative tests often cannot fully replace animal-based data. As a result, international efforts are advancing IATA, which combines multiple data sources to support policy decision-making and ensure environmental safety. This project aims to establish an fish gill cell line acute toxicity test (FCT), develop a fish acute toxicity (AFT) IATA, and gather information on emerging alternative testing methods.The dose-response results from five reference toxicant tests in this project met the standards. Additionally, ten effluent-related substances were assessed to evaluate the applicability of FCT. The findings showed a significant correlation between FCT and AFT, with an r of 0.7732 (p < 0.05). Although sensitivity was lower in the FCT for most substances, 66.7% of the substances had an EC50 within ten times the LC50 of fish acute toxicity tests. The research results from this year will be compiled with international standards to draft a standardized protocol for FCT.Furthermore, the project assessed the correlation and sensitivity of each alternative test and IATA using ten substances. The results confirmed that both alternative tests and IATA are highly correlated with AFT. Due to threshold concentration, IATA predictions of AFT tend to overestimate the toxicity of substances. IATA aids in screening potentially toxic substances while minimizing animal testing, and enhancing environmental protection. In conclusion, the FCT, as an alternative method, shows strong predictive potential. The predictive outcomes of IATA help reduce the underestimation of substance toxicity. Facing developmental challenges, it is recommended to actively adopt emerging alternative testing methods to promote environmental and human health risk assessment and management. | ||
